Directed cell migration is essential for organogenesis. In C. elegans , distal tip cells (DTCs) guide gonadal elongation. While dtc-1 is known, a paralog dtc-2 (ZK123.4) remains uncharacterized. Here, we show that dtc-2 encodes a transmembrane protein expressed in DTCs. Loss of dtc-2 causes gonad migration defects—U-shaped instead of J-shaped turns—and ectopic branching. DTC-2 localizes to focal adhesions and interacts with integrin signaling. Double mutants with dtc-1 exhibit synthetic lethality. Human DTC2 ortholog (TMEM241) rescues migration when expressed in C. elegans , suggesting conserved function. Our work identifies DTC2 as a novel migration regulator with potential implications for metastatic cell invasion.
